Effects of Tolbutamide on Cardiovacular Fine Structures of Alloxan Induced Diabetic Rabbits


Abstract
The author have studied the morphological changes of cardiovascular system induced by oral hypoglycemic agent, D-860, which recently called attention in t:he aspect of its relation to the genesis of hypertension. Alloxan-induced diabetic rabbits were prepared by injecting 130m; of alloxan per kg of body weight. These diabetic animals, then were administered 100mg and 2~Om,g of D-860, per kg of body weights for 3 and 6 months, respectively. Control groups were prepared by the same procedures except giving D-860. Animals were sacrificed and hearts and common carotid arteries were extirpated and then examined by the light and electron microscopes. The results were summarized as follow:
Light microscopically, vacuolar degeneration of myocardial fiber, pericoronary fibrosis, and atrophy of smooth muscle cells and interstitial edema and fibrosi:~ of media in the common carotid artery were observed in both control and experimental groups, but these changes were milder. The changes of the myocardium and coronary arteries were more; severe in control groups, while the changes of the common carotid artery were more severe in the experimental groups.
Electron microscopically, the myocardial cells showed appearance of variable sized vacuoles in `: the sarcoplasm and irregularity of directions of myofibrils. The coronary artery showed increase of collagen fibers under the basement membrane, and common carotid artery showed irregularity of the surface of endothelial cells, edema and increase of collagen fiber in the subintimal layer, thickening- of internal elastic membrane, atrophy of smooth muscle cells, interstitial edema and increase of ~oollagen fiber in the media. Among these changes, those of myocardium and coronary artery were more severe in the control group, but experimental groups showed more severe changes in the common carotid artery.
From the basis that continued administration of D-860 t~ the aloxan-diabetic rabbits could induce more severe pr~aliferative and degenerative changes in carotid artery rather than ma:~cardium and coronary artery, compared with non-D-360-treated control group, the results suggested that these changes of common carotid artery were possibly significant in the genesis of hypertension.
